Genetic Polymorphism Influencing Arsenic Metabolism in Human

چکیده
Groundwater pollution by arsenic is a serious worldwide problem, especially in developing Asian and African countries. Large inter-individual variation in the arsenic metabolism has been reported. Human arsenic (+3 oxidation state) methyltransferase (AS3MT) is known to catalyze the methylation of arsenite. The M287T polymorphism in AS3MT is considered to be related to inter-individual variation in the arsenic metabolism. Several genetic polymorphisms within AS3MT genes in the Japanese, Koreans, Chinese, Mongolians, Uygurs, Tibetans, Vietnamese, Tamangs, Tamils, and Sinhalese in Asia and the Ovambos, Ghanaians, and Xhosas in Africa were investigated. Population differences were observed in the only M287T polymorphism in AS3MT. The Asian populations had 287T variant allele frequencies ranging from 0.000 to 0.041, relatively lower than those of the Africans and Caucasians. The other polymorphisms within AS3MT did not show such ethnic differences. Our findings indicate that the genetic susceptibility to arsenic toxicity in Asian populations is different from that in Africans and Caucasians.

Individual variability in human arsenic metabolism has been reported frequently in the literature. This variability could be an underlying determinant of individual susceptibility to arsenic-induced disease in humans. Recent analysis revealing familial aggregation of arsenic metabolic profiles suggests that genetic factors could underlie interindividual variation in arsenic metabolism. We scree...

We report the association of levels of urinary arsenic (As) compounds (arsenite (AsIII), arsenate (AsV), monomethylarsonic acid (MMAV), dimethylarsinic acid (DMAV), and arsenobetaine (AB)) with genetic polymorphisms of candidate genes, glutathione S-transferase omega 1 (GSTO1) and 2 (GSTO2), mu 1 (GSTM1), and theta 1 (GSTT1), arsenic (+3 oxidation state) methyltransferase (AS3MT), methylenetetr...
متن کاملIndividual Variations in Inorganic Arsenic Metabolism Associated with AS3MT Genetic Polymorphisms
Individual variations in inorganic arsenic metabolism may influence the toxic effects. Arsenic (+3 oxidation state) methyltransferase (AS3MT) that can catalyze the transfer of a methyl group from S-adenosyl-l-methionine (AdoMet) to trivalent arsenical, may play a role in arsenic metabolism in humans. Since the genetic polymorphisms of AS3MT gene may be associated with the susceptibility to inor...
متن کاملMetabolism and toxicity of arsenic: A human carcinogen
Inorganic arsenic is considered the most potential human carcinogen, and humans are exposed to it from soil, water, air and food. In the process of arsenic metabolism, inorganic arsenic is methylated to monomethylarsonic acid and finally to dimethylarsinic acid, followed by excretion through urine. Thus, arsenic exposure may cause DNA hypomethylation due to continuous methyl depletion, facilita...

We report the results of a screen for genetic association with urinary arsenic metabolite levels in three arsenic metabolism candidate genes, PNP, GSTO, and CYT19, in 135 arsenic-exposed subjects from the Yaqui Valley in Sonora, Mexico, who were exposed to drinking water concentrations ranging from 5.5 to 43.3 ppb. We chose 23 polymorphic sites to test in the arsenic-exposed population. Initial...
